| سابقه و هدف: تنش قلیایی با ایجاد pH بالا و تجمع یون سدیم میتواند منجر به کمبود پروتون و تخریب یا مهار شیب/پتانسیل الکتروشیمیایی در سلولهای ریشه شود که در نتیجه جذب آب و یون را مختل میکند. تنش قلیایی فتوسنتز و در نتیجه رشد گیاه را کُند میکند. همچنین قلیائیت باعث تشکیل شکلهای غیر محلول آهن، روی و مس میگردد که قابل جذب برای گیاه نمیباشند، در نتیجه سنتز کلروفیل کاهش یافته و برگهای جوان زرد میشوند. در سالهای اخیر بسیاری از محققین برای کاهش پدیده کمبود سلنیوم در انسان تکنیک غنیسازی زیستی را پیشنهاد میدهند سلنیوم یکی از عناصر غیرضروری ولی سودمند برای گیاهان است که می-تواند عوارض تنش قلیائیت را خنثی یا تا حدودی کاهش دهد. مواد و روشها:. آزمایش بصورت فاکتوریل در قالب طرح پایه بلوکهای کامل تصادفی با سه تکرار در فصل پاییز و زمستان 1402 در مجمتع گلخانهای دانشگاه شهید چمران اهواز اجرا شد. تیمارهای آزمایش شامل غلظتهای مختلف سلنیوم (۰، ۵، ۱۵ و ۳۰ میلیگرم بر لیتر) و سطوح بیکربنات (۰، ۳ و ۶ میلیاکیوالان بر لیتر) بود. بعد از اعمال تیمار و پس از اتمام آزمایش صفات مورفولوژیک مانند ارتفاع نشاء و طول هیپوکوتیل توسط خطکش، قطر هیپوکوتیل توسط کولیس دیجیتالی، وزن تر و خشک شاخساره و ریشه به ترتیب توسط ترازو و خشک کردن در آون 70 درجه سلسیوس به مدت 48 ساعت، سطح برگ توسط دستگاه سطح برگ سنج و صفات فیزیولوژیک نشاء کلمپیچ مانند رنگیزههای فتوسنتزی توسط دستگاه اسپکتروفتومتر، محتوای نسبی آب برگ و نشت الکترولیت با استفاده از معادلات استاندارد اندازهگیری و محاسبه شد. یافتهها: نتایج نشان داد که افزودن سلنیوم به محیط کشت تحت تنش قلیائیت، به طور کلی منجر به افزایش وزن تر و خشک اندامهای مختلف شد. در شرایط تنش، بیشترین وزن تر شاخساره (2 گرم) که از مولفههای یک نشاء با کیفیت میباشد، در تیمار 15 میلیگرم بر لیتر سلنیوم و تنش 3 میلیاکیوالان بر لیتر بیکربنات مشاهده گردید که در قیاس با تیمار صفر میلیگرم بر لیتر سلنیوم و تنش 3 میلیاکیوالان بر لیتر بیکربنات سبب افزایش 27/16 درصدی وزن تر شاخساره نشاء کلم پیچ گردید که از یافتههای جالب توجه این پژوهش میباشد. استفاده از سلنیوم در شرایط تنش قلیائیت باعث کاهش نشت یونی و افزایش محتوای نسبی آب نشاء کلمپیچ شد. نتایج این پژوهش نشان داد که سلنیوم در غلظتهای بهینه میتواند به عنوان یک عامل بهبود دهنده رشد و تحمل گیاه در تنش قلیائیت عمل کند. با این حال، غلظتهای بالای سلنیوم میتواند اثرات سمی داشته باشد و به رشد گیاه آسیب برساند؛ برای نمونه تنها در تیمار بدون تنش، افزایش غلظت سلنیوم تا سطح 30 میلیگرم بر لیتر سبب کاهش 44/59 درصدی وزن تر شاخساره نشاء کلم پیچ در قیاس با تیمار صفر میلیگرم بر لیتر سلنیوم شد. نتیجهگیری: نتایج پژوهشها نشان میدهد که گیاه کلم توانایی غنیسازی سلنیوم را دارد. سطوح مناسب تیمار سلنیوم میتواند باعث بهبود رشد و کیفیت کلم شود. با توجه به مصرف آبهای نامتعارف در گلخانههای تولید نشاء جنوب کشور و ایجاد برگ-های زرد و ساقههای نازک متعاقب استفاده از آن، ارائه راهکار عملی برای غنی سازی کلم پیچ و حل مشکلات واحدهای تولید نشاء کاملاً ضروری بهنظر میرسد. با توجه به تعدیل اثرات نامطلوب کیفیت آب مورد استفاده در گلخانه توسط سطوح بهینه سلنیوم، به نظر میرسد که این پژوهش با اصلاح شیوههای زراعی مرسوم، گامی مهم در جهت توسعه کشاورزی پایدار و افزایش تولید محصولات کشاورزی در مناطق با آبهای قلیایی است. بر اساس نتایج غلظت 15 میلیگرم بر لیتر سلنیوم جهت تعدیل اثرات منفی تنش قلیائیت و بهبود رشد نشاء در گلخانههای تولید نشا کلمپیچ به ویژه در گلخانههای با آبهای قلیایی قابل توصیه است. | ||
