| چکیده | ||
| سابقه و هدف: گیاه دارویی مریم نخودی (Teucrium polium L.) گیاهی چندساله از خانواده نعناع (Lamiaceae) بهخواص کاهندگی قند خون شناخته شده است. شاخههای گلدار آن حاوی ترکیبات فنولی و فلاونوئیدی هستند که اثرات مقوی، ضددرد و آنتیاکسیدانی دارند. با توجه به تأثیر عوامل محیطی بر ترکیبات فیتوشیمیایی، این پژوهش به ارزیابی فعالیت آنتیاکسیدانی، محتوای فنولی و فلاونوئیدی 18 اکوتیپ از جنوب ایران پرداخت. هدف، شناسایی اکوتیپهای برتر با پروفایل بهینه برای کاربردهای دارویی و ارائه پایهای برای حفاظت و تکثیر تجاری آنها بود. مواد و روشها: در این پژوهش 18 اکوتیپ مریم نخودی در مرحله گلدهی کامل از زیستگاههای مختلف در جنوب ایران، شامل استانهای هرمزگان (سیاهو، زیارتعلی، گنو و هماگ)، کرمان (ساردوئیه، مهروئیه، دهبکری، انجرک، ردور، نراب، رمون، راین، کندر، اسفندقه، چاهشاهی و جنتآباد)، بوشهر (کنگان) و کهگیلویه و بویراحمد (دنا) جمعآوری شدند. شاخههای گلدار در سایه و دمای اتاق خشک شده، پودر شدند و عصاره متانولی آنها به روش خیساندن برای ارزیابی خواص فیتوشیمیایی تهیه شد. محتوای فنولی کل با استفاده از معرف فولین- سیوکالتو و اندازهگیری جذب نوری در 750 نانومتر، با کالیبراسیون بر اساس منحنی استاندارد اسید گالیک، تعیین شد. محتوای فلاونوئیدی کل با روش رنگسنجی کلرید آلومینیوم و اندازهگیری جذب نوری در 415 نانومتر، با کالیبراسیون بر اساس منحنی استاندارد کوئرستین، اندازهگیری شد. ظرفیت آنتیاکسیدانی با استفاده از آزمون خنثیسازی رادیکال آزاد 2,2-دیفنیل-1-پیکریلهیدرازیل (DPPH) و اندازهگیری جذب نوری در 517 نانومتر ارزیابی شد. تمامی اندازهگیریها در سه تکرار انجام شد و دادهها با استفاده از تحلیل واریانس یکطرفه (ANOVA) و آزمون چند دامنهای دانکن (05/0>p ) تحلیل شدند. یافتهها: زیستگاه بهطور معنیداری بر ترکیبات فیتوشیمیایی گیاه مریم نخودی تأثیرگذار بود (01/0>p ). در میان ۱۸ اکوتیپ مورد بررسی، اکوتیپ سیاهو (هرمزگان) بیشترین محتوای فنولی (66/86 میلیگرم معادل اسید گالیک بر گرم وزن خشک) و اکوتیپ کندر (کرمان) بیشترین محتوای فلاونوئیدی (45/56 میلیگرم معادل کوئرستین بر گرم وزن خشک) را داشتند. همچنین، اکوتیپ نراب (کرمان) بالاترین مقدار IC50 را (02/85 میکروگرم بر میلیلیتر) نشان داد که این امر بیانگر فعالیت آنتیاکسیدانی ضعیفتر این اکوتیپ نسبت به سایر اکوتیپها است. تحلیل همبستگی نشان داد بین مقدار فنول کل و معکوس مقدار IC50 رابطه مثبت و معنیدار (R² = 0.82) وجود دارد؛ به بیان دیگر، افزایش فنول کل با کاهش مقدار IC50 (و در نتیجه افزایش قدرت آنتیاکسیدانی) ارتباط معنیداری داشت که نقش فنول کل در فعالیت آنتیاکسیدانی گیاه را تأیید میکند. اکوتیپهای دو استان کرمان و هرمزگان بهطور مداوم نسبت به اکوتیپهای استانهای بوشهر و کهگیلویه و بویراحمد عملکرد بهتری داشتند. اکوتیپ سیاهو در محتوای فنولی 25 درصد بالاتر از میانگین سایر اکوتیپها بود؛ در حالیکه کندر در فلاونوئیدها برتری داشت. تفاوتهای آماری معنیدار (05/0>p ) بین اکوتیپها، نشاندهنده تأثیر عوامل محیطی مانند ارتفاع و بارندگی بر تنوع فیتوشیمیایی بود. نتیجهگیری: نتایج این پژوهش نشان داد که شرایط اکولوژیک مناطق مرتفع (از جمله تابش بیشتر، نوسان دمایی و بارندگی کمتر) بهطور قابلتوجهی موجب افزایش ترکیبات فنولی، فلاونوئیدی و ظرفیت آنتیاکسیدانی در گیاه مریم نخودی میشود. این یافتهها لزوم توجه همزمان به عوامل محیطی مانند ارتفاع، دما و بارش را در انتخاب ژرمپلاسمهای برتر برای اهداف دارویی و صنعتی نشان میدهد. معرفی اکوتیپهای پرمحتوا (نظیر کندر، سیاهو و اسفندقه) میتواند زمینهساز توسعهی کشت پایدار، حفظ منابع طبیعی و بهبود کیفیت فرآوردههای دارویی مبتنی بر این گیاه باشد. | ||
